Abstract
This thesis examines modern chromatographic approaches for the isolation, identification, and quantitative determination of phenylethanoid glycosides from plants of the Cistanche genus, with particular attention to echinacoside and acteoside. High-performance liquid chromatography with diode-array detection (HPLC-DAD), ultra-high-performance liquid chromatography with photodiode-array detection (UPLC-PDA), liquid chromatography–tandem mass spectrometry (LC-MS/MS), and high-speed counter-current chromatography (HSCCC) are compared in terms of selectivity, sensitivity, reproducibility, and cost-effectiveness. The results show that HPLC-DAD is the most suitable method for routine quality control and standardization, whereas LC-MS/MS provides the highest analytical certainty at low concentrations. HSCCC is considered the most effective preparative technique for obtaining highly purified echinacoside and acteoside.
